ABSTRACT

BACKGROUND: Aeromonas species are one of the most important etiologies of diseases in fish farms, leading to clinical manifestation and mortality and are associated with public health risks. This study aimed to investigate the prevalence, phenotypic and molecular characteristics of Aeromonas species isolated from farmed Clarias gariepinus using 16 S rRNA sequencing. Additionally, their antibiogram and multiple antibiotic resistance index were determined using a disc diffusion test. RESULTS: A total of 230 Aeromonas strains were isolated from Clarias gariepinus with 40.9% obtained from diseased fish, and 25% isolated from apparently healthy ones. Five different species including Aeromonas caviae, Aeromonas veronii, Aeromonas hydrophila, Aeromonas dhakensis and Aeromonas enteropelogenes were fully identified and genetically characterized. Based on the available literature, this is the first report of Aeromonas enteropelogenes from the study area. The phylogenetic analysis showed genetic heterogeneity and distance within the species and the reference strains. The multiple resistant Aeromonas species were susceptible to ciprofloxacin, gentamycin, and florfenicol. The Aeromonas species' multiple antibiotic resistance index values varied between 0.20 and 0.80 and were isolated from the farms where antibiotics were intensively used. CONCLUSIONS: The diversity of multidrug-resistant Aeromonas species isolated from fish farms is a major threat to fish production giving us more understanding of epidemiology and the multidrug Aeromonas species with a MAR index of greater than 0.2 were isolated from farms where antibiotic use was widespread. As a result, a considerably increased danger of multiple antibiotic resistance spreading to the fish culture environment may impact aquaculture production. Hence there is a need for appropriate and monitored drug usage.

ABSTRACT

BACKGROUND: Exercise-induced oxidative stress is a challenge in equine sports. This study aims at determining the effects of ergothioneine on heat shock protein-70 (HSP-70) following the stress of an endurance exercise of 30 km by Arabian stallions. Molecular docking was also done to investigate the interaction between the ligand ergothioneine and heat shock protein-70 using sulfogalactosylceramide and sulfogalactoglycerolipid as standards. The study involved a total of 18 clinically healthy stallions, with an average age of 6.7 ± 2.4 years and an average weight of 411.54 ± 12.46 kg. Only clinically healthy stallions were selected as subjects. The stallions were divided into two groups of nine stallions each. Group I (ERGX) was administered ergothioneine at a dose of 0.02 mg/kg once daily orally for four weeks while group II (ERGN) was not administered ergothioneine. The activities of the antioxidant enzymes superoxide dismutase, catalase, and glutathione peroxidase were determined in the two groups before and post-exercise. The concentrations of malondialdehyde and HSP-70 were also determined. RESULTS: The results obtained showed that the activities of the antioxidant enzymes and concentration of HSP-70 were higher (P < 0.05) in the ERGX group compared to the ERGN group. The concentration of malondialdehyde was however lower in the ERGX group. Following molecular docking, ergothioneine and the selected standards have common amino acids at the site of interaction with the target protein (HSP-70) suggesting that ergothioneine may have a modulatory effect on the synthesis of HSP-70. CONCLUSION: The results obtained indicated that ergothioneine modulated the synthesis of HSP-70 and the biomarkers of oxidative stress. It was therefore concluded that ergothioneine may be beneficial to horses subjected to endurance exercise.

ABSTRACT

Background and Aim: Heat stress is a major challenge for animals, impairing their welfare and performance. This study aimed to determine the effect of heat stress on the vital and hematobiochemical parameters of healthy dogs. Materials and Methods: The experimental subjects comprised 10 dogs, encompassing seven males and three non-pregnant females between 2 and 3 years of age. Ambient temperature (AT) and relative humidity (RH) were recorded 2 hourly during the day and the temperature humidity index was calculated. Vital parameters [i.e., rectal temperature (RT), respiratory rate, and heart rate (HR)] were assessed and blood was collected from each dog daily for hematobiochemical analysis. Results: The RT (38.5±0.2°C) of dogs exposed to high AT and high RH (HA/HR) conditions was significantly (p<0.05) higher than that of dogs exposed to HA and low RH (LR) conditions (37.2±0.11°C). Under HA/HR conditions, packed cell volume, hemoglobin concentrations, and white blood cell counts were significantly lower than those of the same dogs exposed to HA/LR conditions. Conversely, under HA/HR conditions, the lymphocyte, monocyte, eosinophil, alanine aminotransferase, aspartate aminotransferase, and cortisol values were significantly higher (p<0.05) than the values obtained in dogs exposed to HA/LR conditions. Meanwhile, the alkaline phosphatase, urea, and glucose levels were significantly lower (p<0.05) in dogs exposed to HA/HR conditions. Conclusion: The exposure of healthy dogs to HA/HR conditions induced heat stress, which may have an adverse effect on their immune status, thereby affecting their health and welfare.
